تاثیر بسترهای غیرخاکی بر رشد وشاخص‌های کلنیزاسیون میکوریزی در دو گیاه شبدر و سورگوم

چکیده

اهداف: با توجه به اهمیت بستر کشت گیاه میزبان برای تهیه زادمایه قارچ‌های میکوریز آربوسکولار، در این مطالعه بسترهای غیرخاکی با ویژگی‌های قابل‌دسترس بودن، سبک و ارزان‌قیمت، دارای خواص فیزیکی و شیمیایی مناسب و عدم آلودگی پاتوژنی برای تعیین بستر بهینه گیاه میزبان و تولید زادمایه قارچ، مورد ارزیابی قرار گرفتند.
مواد و روش‌ها: در این آزمایش دو گیاه میزبان شامل شبدر و سورگوم در کشت گلدانی با هفت نوع ترکیب بستر استریل با قارچ AM گونه Diversispora versiformis مایه‌زنی و به مدت چهار ماه در شرایط گلخانه رشد یافتند. تیمارهای آزمایش شامل هفت نوع بستر: خاک شن لومی (LS) و بسترهایی با نسبت حجمی 1:4 کمپوست به پرلیت (CO:P)، کمپوست به ورمیکولیت (CO:VL)، ورمی‌کمپوست به پرلیت (VC:P)، ورمی‌کمپوست به ورمیکولیت (VC:VL)، خاک لوم‌شنی به پرلیت (SL:P)، خاک لوم‌شنی به ورمیکولیت (SL:VL) بودند. آزمایش در قالب کرت خرد شده بر پایه طرح کاملاً تصادفی در سه تکرار انجام شد.
یافته‌ها: بیشترین وزن خشک بخش هوایی و ریشه در هر دو گیاه میزبان در بستر VC:P و VC:VL ثبت شد. غلظت عناصر غذایی اندازه‌گیری شده در هر دو گیاه نیز در بسترهای حاوی ورمی‌کمپوست بیشتر از کمپوست بود. بیشترین درصد کلنیزاسیون طولی ریشه، فراوانی وزیکول و هیف در ریشه و همچنین تعداد اسپور قارچ در بستر، بعنوان شاخص‌های موثر در پتانسیل زاد مایه قارچ، در هر دو گیاه شبدر و سورگوم در بسترهای حاوی کمپوست ( CO:P و CO:VL ) بدست آمد. این شاخص‌ها در بسترهای حاوی ورمی‌کمپوست، کمتر بودند.

چکیده [English]

Background & Objectives: Considering the importance of substrate for host plant culture in production of arbuscular mycorrhizal (AM) fungi inoculum, in this study, non-soil substrates in terms of their accessible, lower weight, inexpensive, appropriate physical and chemical properties with no pathogenic contamination were evaluated to select suitable pot substrate for host plant growth and fungal inoculum production.
Materials & Methods: In a pot culture experiment, clover and sorghum as host plants, were inoculated with the AM fungus Diversispora versiformis and grown in greenhouse conditions for four months with seven types of sterile substrate combinations. Pot substrates were: sandy loam soil (SL), and combinations with 1:4 (v/v) compost:perlite (CO:P), compost:vermiculite (CO:VL), vermicompost:perlite (VC:P), vermicompost:vermiculite (VC:VL), sandy loam:perlite (SL:P), and sandy loam:vermiculite (SL:VL). The experiment was conducted in a split plot format based on a completely randomized design with three replications.
Results: The highest shoot and dry weights of both host plants were recorded in VC:VL and VC:P substrates. Nutrients concentration in both plants were higher in vermicompost than compost based substrates. The highest root length colonization, the frequency of vesicles and hyphae in roots and the number of spores in substrate as efficient indices of fungal inoculum potential were obtained in compost based substrates (CO:P ,CO:VL) for both host plants. These indices were relatively lower in vermicomposr based substrates.
